The objective of the present study was to determine the extent of nicotine uptake in various regions of the brain after systemic delivery. The uptake of [¹⁸F]2-fluoro-2-deoxy-D-glucose (FDG) and [¹⁸F]6-fluoro-L-nicotine (F-LNIC) was determined in mice using in vivo positron emission tomography (PET) and pre-treatment with unlabelled nicotine was also assessed. The uptake of FDG in mice was inhibited by unlabelled nicotine as well as by nicotine administered locally in the brain using intranasal delivery. The uptake of FDG was also inhibited in the presence of nicotine administered via osmotic pumps. Immunohistochemical analyses revealed that nicotine receptors are widely distributed in the brain but are differentially expressed in cell types. Nicotine binds to receptors in regions of the brain that are rich in neuronal cells, including the medial septum, the diagonal band, the corpus striatum and the hippocampus. The exposure of various cell types of the brain, such as microglia and astrocytes, to nicotine may also have implications in drug abuse and the development of addiction.Sosnowiec, Świętokrzyskie Voivodeship
